Effects of Melatonin and/or Magnesium Supplementation in Women with Polycystic Ovary Syndrome

Magnesium and melatonin exert multiple beneficial effects like anti-inflammatory and antioxidant actions. The present study determined the effects of magnesium and/or melatonin supplementation on metabolic profiles in women with polycystic ovary syndrome (PCOS).

The study enrolled 84 subjects with PCOS aged 18–40 years and randomized them to take magnesium, melatonin, magnesium plus melatonin, or a placebo for eight weeks. It drew fasting blood samples at baseline and after the intervention to quantify related variables.

After the 8-week intervention, the study observed-

  • An insignificant marginal difference in waist circumference (WC) between groups.
  • More reductions in hirsutism with Magnesium-melatonin co-supplementation compared with other groups.
  • Significant reduction in serum levels of tumor necrosis factor-α (TNF-α) in the melatonin and co-supplementation groups compared to baseline.
  • More increase in total antioxidant capacity (TAC) levels with magnesium plus melatonin than other treatment groups.

This study shows a favorable effect of co-supplementation of magnesium and melatonin for eight weeks in women with PCOS on hirsutism, serum TNF-α, and TAC levels. Furthermore, the study also shows that melatonin can independently decrease serum values of TNF-α.
